ANXA2 DARPin is a designed ankyrin-repeat protein (DARPin) therapeutic candidate that targets Annexin A2 (ANXA2), a protein found to be highly expressed on the surface of pancreatic cancer stem cells (CSCs). Developed by MedImmune (a subsidiary of AstraZeneca), the drug was identified through a phenotypic selection campaign using a DARPin library. Mechanistically, the ANXA2 DARPin binds to cell-surface ANXA2, leading to a reduction in the frequency of CSCs and inhibiting tumor growth. It also decreases the secretion of pro-tumorigenic cytokines IL-6 and IL-8 and reduces plasmin activation, a pathway linked to the dissemination of cancer cells.
