Overview

ANXV-chemo is a novel protein-drug conjugate (PDC) developed by Annexin Pharmaceuticals, consisting of recombinant human Annexin A5 (ANXV) conjugated to a proprietary chemotherapeutic agent. The drug targets phosphatidylserine (PS), a phospholipid that is typically restricted to the inner leaflet of the cell membrane but becomes abnormally externalized on the surface of tumor cells and tumor-associated vasculature. Annexin A5 binds to PS with high affinity, enabling the targeted delivery and internalization of the cytotoxic payload directly within the tumor microenvironment. Beyond its role as a delivery vehicle, Annexin A5 may also function as an immune checkpoint inhibitor by blocking the immunosuppressive signals generated by externalized PS, such as the promotion of M2 macrophage polarization and T-cell exhaustion. Preclinical studies in triple-negative breast cancer (TNBC) models have demonstrated that ANXV-chemo is significantly more cytotoxic than the free chemotherapy agent and can lead to rapid tumor volume reduction and vascular necrosis.
