Anzurstobart (CC-95251) is a humanized monoclonal antibody targeting Signal Regulatory Protein alpha (SIRPα), developed by Celgene (a subsidiary of Bristol Myers Squibb). SIRPα is an inhibitory receptor expressed on myeloid cells, including macrophages and dendritic cells. By binding to SIRPα, anzurstobart disrupts its interaction with CD47, a protein often overexpressed on cancer cells that sends a "don't eat me" signal to immune cells. This blockade promotes the phagocytosis of tumor cells by macrophages and enhances the adaptive immune response. It is being investigated as a monotherapy and in combination with other agents like cetuximab or rituximab for the treatment of advanced solid tumors (such as colorectal cancer and squamous cell carcinoma of the head and neck) and hematologic malignancies (including diffuse large B-cell lymphoma and follicular lymphoma).
