Overview

AO-176 is a humanized IgG2 monoclonal antibody that specifically targets CD47, a cell surface protein overexpressed on many tumor types. By binding to CD47, AO-176 blocks its interaction with signal regulatory protein alpha (SIRPα) on phagocytes, thereby disrupting the "don't eat me" signal that allows cancer cells to evade immune clearance. The drug promotes phagocytosis of tumor cells, induces direct cytotoxic cell death through programmed cell death type III and immunogenic cell death by releasing damage-associated molecular patterns (DAMPs), and preferentially binds to tumor cells over normal cells due to enhanced affinity in acidic microenvironments typical of tumors. Notably, AO-176 exhibits negligible binding to red blood cells, reducing the risk of hemagglutination and anemia seen with other anti-CD47 agents. It is being developed as both monotherapy and in combination regimens for relapsed/refractory multiple myeloma and various advanced solid tumors[1][2][4][5][6].
