AOB01-0036 is an orally bioavailable small molecule inhibitor of Son of Sevenless homolog 1 (SOS1), developed by Double Wings Original (Shanghai) Pharmaceutical. SOS1 acts as a guanine nucleotide exchange factor (GEF) that facilitates the conversion of KRAS from its inactive GDP-bound form to its active GTP-bound form. By binding to the catalytic domain of SOS1, AOB01-0036 prevents the activation of KRAS and downstream MAPK signaling pathways. This "pan-KRAS" approach is intended to treat advanced solid tumors harboring various KRAS mutations, potentially overcoming resistance to allele-specific KRAS inhibitors. It is currently being evaluated in Phase I clinical trials for patients with advanced, unresectable, or metastatic solid tumors.
