Overview

AP-144 + Keytruda is a combination immunotherapy regimen being developed by Alphageneron for the treatment of Stage IV non-small cell lung cancer (NSCLC). The regimen combines AP-144, an autologous cell therapy comprising natural killer (NK) cells that have been activated ex vivo to target membrane-bound Heat Shock Protein 70 (Hsp70), with Keytruda (pembrolizumab), a programmed death receptor-1 (PD-1) blocking monoclonal antibody. Hsp70 is frequently overexpressed on the surface of tumor cells, providing a specific target for the cytotoxic NK cells. By combining this targeted cell therapy with pembrolizumab-mediated checkpoint blockade, the treatment aims to overcome tumor-induced immunosuppression and promote a more robust and sustained anti-cancer immune response. The combination is currently being evaluated in Phase 2 clinical trials.
