AP-161 + Keytruda is a combination therapy being developed by Alphageneron for the treatment of metastatic Stage IV non-small cell lung cancer (NSCLC). The regimen combines AP-161, which consists of allogeneic expanded natural killer (NK) cells targeted toward membrane-bound Heat Shock Protein 70 (mHsp70), with Keytruda (pembrolizumab), a programmed death receptor-1 (PD-1) blocking monoclonal antibody. mHsp70 is a tumor-specific antigen overexpressed on the surface of various cancer cells, making it a viable target for NK cell-mediated cytotoxicity. By combining these targeted NK cells with pembrolizumab-mediated checkpoint blockade, the therapy aims to overcome tumor-induced immunosuppression and promote a more robust immune attack against metastatic lesions. The program is currently in Phase 1 clinical trials.
