Overview

AP-325 (laflunimus) is a novel, non-opioid small molecule drug candidate developed for the treatment of neuropathic pain. It acts as a positive allosteric modulator of the GABA A receptor, specifically targeting GABAergic signaling in the dorsal root ganglion (DRG). By enhancing inhibitory neurotransmission via GABA A receptors, AP-325 counteracts excessive neuronal excitation associated with chronic neuropathic pain. In clinical studies, AP-325 demonstrated rapid onset and long-lasting reduction in pain intensity without central side effects such as sedation or dizziness. The drug has shown significant improvements in sleep quality and reductions in anxiety and depression symptoms among patients with peripheral post-surgical neuropathic pain. Its safety profile is comparable to placebo, and it does not carry risks of dependence typical of opioid therapies[5][6][7][8].
