AP-401 is an allogeneic, "off-the-shelf" natural killer (NK) cell therapy developed by Alphageneron. It is derived from induced pluripotent stem cells (iPSCs) and engineered to express a bi-specific chimeric antigen receptor (CAR) targeting CD155 (Poliovirus receptor) and CD73 (5'-nucleotidase). These two targets are frequently overexpressed in various solid tumors and play critical roles in immune evasion and the maintenance of an immunosuppressive tumor microenvironment. By targeting both antigens simultaneously, AP-401 is designed to enhance tumor recognition and overcome resistance mechanisms common in solid malignancies. Preclinical studies in animal models have demonstrated promising efficacy, and the therapy represents a scalable approach to cell-based cancer treatment.
