AP-5 (also known as APV) is a potent and selective competitive antagonist of the N-methyl-D-aspartate (NMDA) receptor. It functions by binding to the glutamate recognition site of the NMDA receptor complex, effectively blocking the action of the excitatory neurotransmitter glutamate. AP-5 is a cornerstone tool in neuroscience research, extensively utilized to study the mechanisms of synaptic plasticity, such as long-term potentiation (LTP) and long-term depression (LTD), which are cellular correlates of learning and memory. Although it has been used in preclinical animal models to explore gating deficits related to schizophrenia and other neuropsychiatric disorders, it is not used clinically in humans due to its poor blood-brain barrier permeability and potential for dissociative side effects.
