AP052 is a **novel tubulin inhibitor payload** designed as the cytotoxic component of next-generation antibody-drug conjugates (ADCs), most notably in ADRX-0706, developed by Adcentrx Therapeutics. It is conjugated to monoclonal antibodies via a proprietary cleavable linker using Adcentrx's i-Conjugation technology, resulting in selective delivery of AP052 to tumor cells expressing targets such as Nectin-4. Upon internalization, AP052 binds to tubulin, **inhibiting its polymerization** to cause G2/M cell cycle arrest and subsequently induce apoptosis. AP052 and its ADCs offer improved pharmacokinetics, a widened therapeutic window, minimal free payload in plasma, and a reduced risk of peripheral neuropathy compared to other tubulin inhibitor-based ADC payloads[2][5][6][8][10].
