AP13 (also known as APL-9796) is a first-in-class monoclonal antibody (mAb) designed to target the zinc transporter protein ZIP12 (SLC39A12). Developed by Apollo Therapeutics in collaboration with Imperial College London, AP13 is being investigated as a disease-modifying therapeutic for pulmonary hypertension (PH), including WHO Group 1 Pulmonary Arterial Hypertension (PAH) and WHO Group 3 PH associated with interstitial lung disease (PH-ILD). ZIP12 is a protein that regulates intracellular zinc levels and has been found to be significantly upregulated in the lungs of patients with pulmonary hypertension, where it contributes to pulmonary vascular remodeling. By inhibiting ZIP12, AP13 aims to reverse or prevent this remodeling process. The drug is administered via subcutaneous injection and has received Orphan Drug Designation from the US FDA. It has progressed from Phase 1 studies in healthy volunteers to Phase 2 clinical trials in patients with pulmonary hypertension.
