AP21163

AP21163 is a small molecule tyrosine kinase inhibitor (TKI) developed by ARIAD Pharmaceuticals as part of a research program to identify potent inhibitors of the BCR-ABL fusion protein. It was specifically engineered to overcome the T315I gatekeeper mutation, which renders chronic myeloid leukemia (CML) resistant to first- and second-generation TKIs like imatinib, nilotinib, and dasatinib. AP21163 demonstrated high potency against both wild-type and mutant BCR-ABL in preclinical studies. Although it showed promising activity, ARIAD ultimately advanced a related but optimized analog, AP24534 (ponatinib), into clinical trials and eventual regulatory approval. Consequently, development of AP21163 as a standalone therapeutic was discontinued.
