Overview

AP31969 is a second-generation, oral small molecule inhibitor of small-conductance calcium-activated potassium (SK) channels, developed by Acesion Pharma for the chronic maintenance treatment of atrial fibrillation (AF). It is designed to prevent AF recurrence by selectively inhibiting SK channels in the atria, thereby reducing proarrhythmia risk—a major limitation of existing antiarrhythmic drugs. Preclinical studies have shown AP31969 has a favorable cardiac safety profile with no QT interval prolongation and low risk of drug-drug interactions. The drug demonstrates good oral pharmacokinetics across animal models and pronounced antiarrhythmic effects in the atria while sparing ventricular tissue. As of 2024, AP31969 is undergoing Phase 1 clinical trials for AF[1][4][5][7].
