Overview

AP5280 is a novel polymer-conjugated platinum-based chemotherapeutic agent developed for the treatment of solid tumors. It consists of a platinum (Pt) moiety covalently linked to the water-soluble, biocompatible copolymer N-(2-hydroxypropyl)methacrylamide (pHPMA), designed to enhance tumor accumulation via the enhanced permeability and retention effect. This formulation aims to increase therapeutic efficacy while reducing systemic toxicity compared to traditional platinum drugs like cisplatin and carboplatin. The drug acts as a DNA alkylating agent, forming DNA adducts that inhibit DNA replication and transcription, ultimately leading to cancer cell death. Clinical studies have shown that AP5280 allows higher dosing with reduced renal toxicity and myelosuppression relative to conventional platinum agents[1][5][6].
