Overview

A combination of **apalutamide** and **enzalutamide**, both of which are next-generation androgen receptor inhibitors, each approved for the treatment of metastatic castration-sensitive prostate cancer (mCSPC) and non-metastatic castration-resistant prostate cancer (nmCRPC) in combination with androgen deprivation therapy (ADT). Both drugs act as antagonists of the androgen receptor, inhibiting the growth-promoting effects of androgens on prostate cancer cells. There is currently no clinical evidence or regulatory approval for the concurrent use of apalutamide and enzalutamide in a single regimen; instead, these drugs are used as alternatives to one another, not together, as both agents target the same molecular pathway and combination may increase toxicity without proven added benefit. Clinical comparisons suggest subtle differences in efficacy and side effect profiles, but their combination as a regimen is not described in medical literature or guidelines[1][2][3][4][5][6][9][13].
