This combination therapy consists of apatinib, a selective vascular endothelial growth factor receptor-2 (VEGFR2) tyrosine kinase inhibitor, dalpiciclib (SHR6390), a selective cyclin-dependent kinase 4 and 6 (CDK4/6) inhibitor, and endocrine therapy (such as aromatase inhibitors or fulvestrant). Developed in collaboration with Fudan University and Jiangsu Hengrui Pharmaceuticals, this regimen is being evaluated as a first-line treatment for patients with hormone receptor-positive (HR+), human epidermal growth factor receptor 2-negative (HER2-) advanced breast cancer, specifically the SNF4 molecular subtype. The SNF4 subtype is characterized by high expression of angiogenesis-related genes, providing a rationale for adding the anti-angiogenic agent apatinib to the standard-of-care CDK4/6 inhibitor and endocrine therapy backbone to improve clinical outcomes and overcome resistance.
