This is a chemo-immunotherapy combination regimen being investigated for the perioperative treatment of locally advanced gastric adenocarcinoma and gastroesophageal junction (GEJ) adenocarcinoma. It combines the FLOT chemotherapy backbone (docetaxel, oxaliplatin, fluorouracil, and leucovorin) with sintilimab, a PD-1 monoclonal antibody, and apatinib, a small-molecule tyrosine kinase inhibitor targeting VEGFR-2. The rationale is to combine the cytotoxic effects of chemotherapy with the immune checkpoint inhibition of sintilimab and the anti-angiogenic properties of apatinib to improve pathological complete response (pCR) rates and survival outcomes.
