This combination therapy consists of apatinib (apatinib mesylate), doxorubicin, and ifosfamide, and is being investigated for the treatment of advanced or metastatic soft tissue sarcoma (STS). Apatinib is a small molecule tyrosine kinase inhibitor that selectively targets vascular endothelial growth factor receptor-2 (VEGFR-2), thereby inhibiting tumor angiogenesis. Doxorubicin is an anthracycline antibiotic that acts as a cytotoxic agent by intercalating into DNA and inhibiting topoisomerase II, leading to DNA damage and cell death. Ifosfamide is a nitrogen mustard alkylating agent that cross-links DNA, interfering with replication and transcription. This triplet regimen, notably studied at Sun Yat-sen University Cancer Center, aims to enhance therapeutic efficacy in sarcoma subtypes by combining targeted anti-angiogenic effects with standard cytotoxic chemotherapy.
