Apatinib + FOLFOX is a combination regimen used primarily in the treatment of advanced or metastatic colorectal cancer and has also been studied in gastric cancer. Apatinib is a small molecule tyrosine kinase inhibitor that selectively targets vascular endothelial growth factor receptor 2 (VEGFR-2), thereby inhibiting tumor angiogenesis and promoting apoptosis and autophagy in tumor cells. FOLFOX is a chemotherapy regimen consisting of folinic acid (leucovorin), fluorouracil (5-FU), and oxaliplatin. These agents work synergistically to destroy rapidly dividing cancer cells through DNA damage and inhibition of DNA synthesis. The combination aims to enhance antitumor efficacy by targeting both angiogenesis pathways (via apatinib) and cell proliferation pathways (via cytotoxic chemotherapy)[1][3][6].
