Drug intelligence / Profile preview
APC-inhibitory peptide is an experimental peptide-based therapeutic designed to selectively inhibit the anticoagulant activity of activated protein C (APC). In hemophilia, recurrent joint bleeding (hemarthrosis) leads to hemophilic arthropathy, a progressive joint disease driven by impaired thrombin generation and chronic inflammation. APC, which is abundant in hemophilic joints, downregulates thrombin generation by inactivating coagulation factors Va and VIIIa, and may also exacerbate cartilage degradation. By systemically inhibiting APC's anticoagulant function, the APC-inhibitory peptide aims to rebalance coagulation, enhance thrombin generation, and reduce intra-articular bleeding, potentially offering a non-factor replacement strategy for joint protection in hemophilia.
