Apc001 is a nucleic acid aptamer drug developed as a sclerostin inhibitor for bone anabolic therapies. It was designed by modifying the aptamer aptscl56 to improve its half-life and stability. Mechanistically, it targets the "loop3 domain" of sclerostin (SOST), inhibiting its function and thereby promoting bone formation without increasing cardiovascular risk. Preclinical studies have shown that Apc001 increases bone mass, improves microarchitecture integrity, and enhances mechanical properties in animal models of osteoporosis and osteogenesis imperfecta. The drug has received orphan drug designation from the US FDA for osteogenesis imperfecta and rare pediatric disease designation for X-linked hypophosphatemia (XLH). Clinical trials are scheduled to begin in 2024[2][4][5].
