APC148 is a first-in-class, selective zinc-chelating β-lactamase inhibitor developed to combat antimicrobial resistance in Gram-negative bacteria. Its unique mechanism of action targets and inhibits bacterial metallo-β-lactamase (MBL) enzymes—key mediators of resistance to beta-lactam antibiotics—thereby restoring the efficacy of these antibiotics against multidrug-resistant pathogens. Unlike existing serine-β-lactamase (SBL) inhibitors, APC148 specifically addresses MBLs for which there are currently no approved inhibitors on the market. The drug is being developed as an intravenous adjuvant therapy to be combined with reserve group antibacterials such as carbapenems and cephalosporins for the treatment of complicated Gram-negative infections. Clinical development is led by AdjuTec Pharma, with phase 1 trials underway in healthy volunteers[1][2][3][5][6][7][8].
