APC201 is a topical small molecule formulation of the non-steroidal anti-inflammatory drug (NSAID) diclofenac, developed by Andros Pharmaceuticals for the treatment of pain associated with knee osteoarthritis. It functions by inhibiting the cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2) enzymes, which are responsible for the production of prostaglandins that mediate inflammation and pain. The drug utilizes a proprietary, patented dual-action lipid formulation that allows for a 4% concentration of the active ingredient, significantly higher than the 2% concentration found in marketed comparators like Pennsaid. This formulation is delivered as a foam and is designed to enhance drug accumulation in the skin and penetration into the affected joint without the use of irritating solvents such as dimethyl sulfoxide (DMSO). APC201 has demonstrated positive results in Phase I/IIa clinical trials, showing improved pain reduction and better skin tolerability compared to existing topical NSAID treatments.
