APD-IL10 is a preclinical-stage bifunctional fusion protein developed by Protheragen for the treatment of solid tumors. The molecule is designed to simultaneously target the Programmed Cell Death Protein 1 (PD-1) receptor and the Interleukin-10 (IL-10) pathway. It typically consists of a PD-1 antagonist (such as an anti-PD-1 antibody or antibody fragment) fused to IL-10. This dual-action approach aims to overcome immune suppression in the tumor microenvironment by blocking the inhibitory PD-1/PD-L1 checkpoint while simultaneously delivering IL-10 signaling to tumor-infiltrating lymphocytes. Although IL-10 is traditionally characterized as an anti-inflammatory cytokine, in the context of oncology, it has been shown to promote the expansion and effector function of CD8+ T cells. By co-localizing these two immune-modulating signals, APD-IL10 is intended to enhance anti-tumor efficacy and potentially overcome resistance to standard checkpoint inhibitor monotherapies.
