Overview

aPD1-MSLN-CAR T cells are an investigational autologous chimeric antigen receptor (CAR) T cell therapy engineered to target mesothelin (MSLN), an antigen overexpressed in various solid tumors. These CAR T cells are further modified to express or secrete an anti-PD-1 nanobody or antibody, which blocks the programmed cell death protein 1 (PD-1) pathway on immune and tumor cells. This dual mechanism aims to enhance antitumor activity by both directly targeting MSLN-positive cancer cells and overcoming local immunosuppression mediated by the PD-1/PD-L1 axis. The therapy is being evaluated for safety and efficacy in patients with advanced solid tumors expressing mesothelin[2][3][4][6].
