APG-1842 is a small molecule, covalent inhibitor specifically targeting the KRAS G12C mutant protein. It acts by irreversibly binding to the inactive (GDP-bound) form of KRAS G12C, thereby blocking downstream signaling pathways that drive tumor growth in cancers harboring this mutation. Developed by Ascentage Pharma, APG-1842 was investigated for its potential in treating solid tumors with KRAS G12C mutations, including non-small cell lung cancer (NSCLC), colorectal cancer, and gastric cancer. The drug demonstrated high selectivity and potency in preclinical studies but has not advanced beyond preclinical development[1][3][7][9].
