Overview

APG-265 is a preclinical proteolysis-targeting chimera (PROTAC) MDM2 degrader being developed by Ascentage Pharma to reactivate the p53 tumor suppressor pathway in cancer. By inducing ubiquitination and proteasomal degradation of the E3 ligase MDM2, APG-265 is designed to relieve negative regulation of wild-type p53, thereby promoting p53-dependent cell-cycle arrest and apoptosis in tumor cells with intact TP53. APG-265 is part of Ascentage Pharma’s apoptosis-focused oncology pipeline and represents a targeted, mechanism-based antineoplastic agent for hematologic malignancies and solid tumors characterized by functional p53 and MDM2 dependence.[3][6][7][9]
