Overview

APG Tn-9mer is a therapeutic cancer vaccine candidate developed by Vaxil Bio. It is a synthetic glycopeptide consisting of a 9-amino acid peptide derived from the signal (leader) sequence of the MUC1 protein, which is glycosylated with the Tn antigen (N-acetylgalactosamine). MUC1 is a transmembrane glycoprotein that is overexpressed and aberrantly glycosylated in many cancers, including multiple myeloma and various solid tumors. Unlike traditional MUC1 vaccines that target the tandem repeat domain, APG Tn-9mer targets the highly conserved leader sequence, which is essential for protein trafficking and is present in all MUC1 isoforms. By incorporating the tumor-associated Tn carbohydrate, the vaccine is designed to induce a robust T-cell mediated immune response against malignant cells while overcoming the immune tolerance often associated with native MUC1 structures.
