APG777 is a novel, subcutaneous, extended half-life humanized IgG1 monoclonal antibody specifically engineered to bind with high affinity to interleukin-13 (IL-13), a critical cytokine involved in the pathogenesis of atopic dermatitis and other inflammatory diseases. By inhibiting IL-13 signaling, APG777 interrupts downstream inflammatory pathways implicated in moderate-to-severe atopic dermatitis. The drug features an amino acid modification in its Fc region that extends its half-life (approximately 75–77 days), enabling less frequent dosing—potentially as infrequently as once every three to twelve months. APG777 is being developed by Apogee Therapeutics and represents their lead clinical-stage product candidate from a collaboration with Paragon Therapeutics. It is currently in phase 2 clinical trials for atopic dermatitis, with additional development planned for asthma and eosinophilic esophagitis[2][3][4][5][6][7].
