Overview

APG990 is a novel, subcutaneous, half-life-extended monoclonal antibody that targets OX40 ligand (OX40L), also known as tumor necrosis factor ligand superfamily member 4. By binding to OX40L and blocking its interaction with the OX40 receptor, APG990 modulates immune responses upstream in the inflammatory cascade. This mechanism allows for broad inhibition of Type 1, Type 2, and Type 3 inflammation pathways. The drug has demonstrated an extended pharmacokinetic profile with a half-life of approximately 60 days in Phase 1 studies and was well tolerated at doses up to 1,200 mg. Its primary indication is atopic dermatitis (AD), where it is being developed as both a monotherapy and as part of a coformulated combination (APG279) with another monoclonal antibody (APG777). The extended half-life supports maintenance dosing every three to six months[1][5][6][8].
