APH-0911 is an oral nanotechnology formulation of the taxoid prodrug 10-α-GAG-DT, designed to improve the oral bioavailability of docetaxel. Developed by Aphios Corporation, the drug utilizes biodegradable PLGA (poly lactic-co-glycolic acid) polymer nanospheres manufactured using proprietary SuperFluids™ PNS technology. After oral administration and absorption in the small intestine, serum enzymes cleave the prodrug to release bioactive docetaxel. Docetaxel acts by binding to tubulin and stabilizing microtubules, which prevents their depolymerization and leads to cell cycle arrest and apoptosis. APH-0911 is currently in preclinical development for the treatment of various cancers, including breast and ovarian cancer, through low-dosage oral administration.
