APH-0912 is a monoclonal antibody-drug conjugate (ADC) being developed by Aphios Corporation for the treatment of metastatic breast cancer and other metastatic malignancies. The drug consists of a tumor-specific monoclonal antibody covalently linked to a docetaxel (Taxotere) prodrug. To enhance water solubility, hydrophilic sugar molecules are tethered to the C-10 hydroxyl group of the taxoid payload via an ester linkage. Upon intravenous administration, the antibody component targets specific tumor-associated antigens on cancer cells. Serum enzymes then cleave the prodrug, releasing the bioactive anti-mitotic agent docetaxel directly at the tumor site. Preclinical studies have demonstrated that these docetaxel prodrug derivatives exhibit dose-dependent activity superior to docetaxel alone in certain cancer models.
