Overview

APIO-EE-9 is a novel small molecule antagonist of Aurora kinase A and B. It functions by binding to the ATP-binding pocket of these kinases, thereby inhibiting their enzymatic activity. In preclinical studies, APIO-EE-9 has demonstrated the ability to suppress the growth and colony formation of esophageal cancer cells and induce apoptosis. It also reduces the phosphorylation of histone H3 (Ser10), a downstream marker of Aurora kinase activity, leading to mitotic defects such as the formation of multiple nuclei and centrosomes. Research conducted at Zhengzhou University Hospital has shown that APIO-EE-9 significantly decreases tumor size in esophageal patient-derived xenograft (PDX) models, suggesting its potential as a therapeutic agent for esophageal cancer.
