Overview

APL-030 is a novel, selective, and orally bioavailable small molecule inhibitor of General Control Nondenressible 2 (GCN2), also known as EIF2AK4. GCN2 is an evolutionarily conserved kinase that serves as a pivotal regulator of the Integrated Stress Response (ISR), an adaptive cellular program typically triggered by amino acid scarcity. By acting as an ATP-competitive inhibitor, APL-030 prevents the GCN2-mediated phosphorylation of the translation initiation factor eIF2α, thereby blocking the ISR-driven survival program that cancer cells utilize to adapt to nutrient-poor microenvironments. Developed by Apollo Therapeutics, APL-030 has demonstrated potent preclinical efficacy in acute myeloid leukemia (AML) models, including patient-derived samples and leukemic stem cell-enriched populations, where it induces caspase-dependent apoptosis and inhibits tumor growth. Based on these results, clinical development is planned for hematological tumors.
