Overview

APL 400-047 is a DNA-based vaccine candidate developed by Apollon, Inc. (later acquired by Wyeth) for the prevention of HIV-1 infection. The vaccine consists of a plasmid DNA encoding the HIV-1 Gag and Pol proteins. It is designed to stimulate both humoral (antibody) and cellular (T-cell) immune responses by mimicking a live-attenuated virus infection without the associated risks of using a live pathogen. In clinical trials, it was evaluated for safety and immunogenicity when administered intramuscularly via traditional needle and syringe or the Biojector 2000 needle-free jet injection system. The program was part of early efforts in the 1990s to develop DNA vaccines for HIV, but development did not progress to late-stage efficacy trials.
