Aplindore is a small molecule dopamine receptor partial agonist that was primarily investigated for the treatment of Parkinson's disease and Restless Legs Syndrome (RLS). It acts as a high-affinity partial agonist at the dopamine D2 and D3 receptors. By modulating dopamine activity rather than fully activating or blocking the receptors, aplindore aims to provide symptomatic relief with a reduced risk of side effects like dyskinesia or dopamine dysregulation syndrome compared to full agonists. The drug was originally developed by Wyeth and later licensed to Neurogen Corporation for further clinical evaluation. Development was ultimately discontinued following Phase 2 clinical trials.
