Overview

**APMV-4** is a naturally occurring avian paramyxovirus serotype 4, identified as a novel oncolytic virus (OV) with superior antitumor activity compared to Newcastle disease virus (NDV/APMV-1) strains like LaSota in preclinical models. It selectively replicates in and lyses cancer cells due to their defective antiviral responses, while sparing normal cells, and is not a human pathogen with no pre-existing immunity in humans. Intratumoral administration in syngeneic mouse models of B16F10 melanoma and CT26.WT colon carcinoma demonstrated potent tumor growth inhibition, complete remissions (up to 100% when combined with VEGF-C expression), extended survival, and long-term protection against tumor rechallenge, correlated with enhanced T-cell and NK-cell infiltration and lymphangiogenesis. Recombinant versions (rAPMV-4) have been engineered via reverse genetics to express transgenes like GFP, enabling further optimization for therapeutic delivery.[1][2][5][7]
