APO010

APO010 is a first-in-class investigational anticancer agent that is a multimeric (hexameric) recombinant form of the human Fas ligand (also known as CD95 ligand). It acts as an apoptosis stimulant by specifically binding to the Fas receptor (CD95) on cell surfaces and inducing programmed cell death via the extrinsic apoptotic pathway. This mechanism targets malignant cells for destruction and is independent of the cell cycle. APO010 has demonstrated preclinical efficacy in multiple cancer types including multiple myeloma, glioblastoma stem-like cells, non-small cell lung cancer, and ovarian cancer. It does not cross-react with known multidrug resistance mechanisms and may act synergistically with other anticancer agents such as checkpoint inhibitors. The drug was originally developed by Apoxis and further developed by Allarity Therapeutics (formerly Oncology Venture A/S) and Valerio Therapeutics.
