apoA-I adenovirus is a **recombinant oncolytic adenovirus vector engineered to express the human apolipoprotein A1 (ApoA-I) gene**. It is designed as a gene therapy to deliver and express ApoA-I in target tissues. ApoA-I is the main protein component of high-density lipoprotein (HDL) and mediates cholesterol efflux, helping to regulate cholesterol homeostasis. In preclinical cancer studies, the vector showed the ability to promote cholesterol efflux, inhibit tumor growth and metastasis (notably in triple-negative breast cancer), and prolong survival in animal models. Mechanistically, it acts via the cholesterol/IKBKB/FOXO3a/KRT14 pathway to downregulate pro-metastatic processes. Safety studies in animals demonstrated tolerability at high doses.
