ApoB ATAC is an investigational oral small molecule heterobifunctional protein degrader being developed by Avilar Therapeutics for the treatment of atherosclerotic cardiovascular disease (ASCVD). The drug is part of Avilar's ATAC (ASGPR-Targeting Chimera) platform, which is designed to degrade extracellular and circulating proteins. ApoB ATAC works by simultaneously binding to Apolipoprotein B (ApoB)—the primary protein component of pro-atherogenic lipoproteins such as LDL and VLDL—and the asialoglycoprotein receptor (ASGPR), a liver-specific endocytic receptor. This linkage triggers the internalization of the ApoB-ATAC-ASGPR complex into hepatocytes, where the target protein is trafficked to the lysosome for degradation. By reducing the levels of circulating ApoB, the therapy aims to lower atherogenic lipid levels and mitigate cardiovascular risk.
