Overview

apolipoprotein A-I biovector is a therapeutic candidate developed by Biovector Therapeutics (later acquired by BioAlliance Pharma) that utilizes supramolecular biovector (SMBV) technology to deliver apolipoprotein A-I (ApoA-I). SMBVs are nanoparticle delivery systems typically composed of a cross-linked polysaccharide core surrounded by a phospholipid bilayer, designed to mimic the structure of natural lipoproteins to improve the stability and delivery of proteins. The drug was primarily investigated for the treatment of atherosclerosis and hypercholesterolemia. Its mechanism of action involves the delivery of functional ApoA-I to the plasma, where it serves as the primary acceptor for cholesterol efflux from peripheral tissues, particularly macrophages in arterial walls, via the ATP-binding cassette transporter A1 (ABCA1). This process, known as reverse cholesterol transport, facilitates the transport of cholesterol back to the liver for excretion, thereby potentially promoting the regression of atherosclerotic plaques.
