Overview

Apolizumab is a humanized monoclonal IgG1 antibody that targets the 1D10 antigen, a polymorphic determinant on the beta chain of HLA-DR, which is expressed on normal and neoplastic B cells as well as some other immune cells. By binding to this target, apolizumab induces complement-mediated cytotoxicity, antibody-dependent cell-mediated cytotoxicity (ADCC), and direct apoptosis in 1D10-positive B cells. It was developed primarily for hematologic malignancies such as chronic lymphocytic leukemia (CLL), non-Hodgkin lymphoma (NHL), and acute lymphoblastic leukemia (ALL). Clinical trials also explored its use in solid tumors. The drug showed some clinical activity but development was discontinued due to limited efficacy and dose-limiting toxicities including infusion reactions, aseptic meningitis, and hemolytic uremia[1][2][3][4][7].
