APP1-Tau1 Rigid is a synthetic **conjugate peptide** composed of two sequences—the APP1 fragment (amino acids 390–412) of the amyloid precursor protein and the Tau1 fragment (amino acids 19–34) of tau protein—linked by a **rigid peptide bridge**. Designed to inhibit pathological binding between APP and Tau, which plays a role in the development of Alzheimer's disease, this molecule shows the ability to block the interaction in vitro and reduce amyloid-beta plaque formation and cognitive decline in preclinical Alzheimer’s disease models. Both nasal and oral administration of the rigid-linked peptide demonstrated improvement in cognitive function and a reduction in pathological markers in 5xFAD transgenic mice, indicating therapeutic potential as a disease-modifying agent targeting the APP-Tau interaction in Alzheimer’s disease[1][4][7].
