Drug intelligence / Profile preview
APR-1051 + Keytruda (pembrolizumab) is an investigational combination therapy consisting of APR-1051, a potent and highly selective oral small molecule inhibitor of WEE1 kinase, and Keytruda, an FDA-approved anti-PD-1 monoclonal antibody. Developed by Aprea Therapeutics, APR-1051 is designed to target the DNA damage response (DDR) pathway, promoting premature mitosis and cell death in biomarker-defined cancers, particularly those with Cyclin E overexpression or HPV-positive status. Keytruda (pembrolizumab), developed by Merck & Co., is an immune checkpoint inhibitor that blocks the interaction between PD-1 and its ligands, enhancing T-cell-mediated anti-tumor immune responses. The combination is being preclinically and clinically evaluated for the treatment of HPV-positive head and neck squamous cell carcinoma (HNSCC), leveraging the synergistic potential of WEE1-mediated replication stress induction and immune checkpoint blockade.
