Overview

The combination of aprinocarsen, carboplatin, and gemcitabine is an investigational regimen for cancer treatment. **Carboplatin** is a platinum-based chemotherapy agent that forms DNA crosslinks, inhibiting DNA replication and transcription. **Gemcitabine** is a nucleoside analog that inhibits DNA synthesis by incorporating into DNA and RNA chains, leading to chain termination. **Aprinocarsen** (also known as ISIS 3521) is an antisense oligonucleotide targeting protein kinase C-alpha (PKC-α), designed to inhibit the expression of PKC-α mRNA, thereby potentially reducing tumor cell proliferation and survival. This combination has been explored in clinical trials for advanced cancers such as non-small cell lung cancer (NSCLC) or other solid tumors where platinum-based doublets are standard therapy[2][3]. The rationale for combining these agents includes targeting multiple pathways involved in tumor growth and resistance.
