Overview

Aprutumab (BAY 1179470) is a fully human monoclonal antibody that targets fibroblast growth factor receptor 2 (FGFR2), specifically binding to the FGFR2 isoforms IIIb and IIIc. It was developed as an antineoplastic agent with potential activity against various solid tumors by antagonizing the FGFR2 pathway. Aprutumab was also used as the antibody component in the antibody-drug conjugate aprutumab ixadotin (BAY 1187982), which linked it to an auristatin-based cytotoxic payload for targeted cancer therapy. The drug was developed by Bayer and reached phase I clinical trials for advanced refractory solid tumors but did not progress further due to dose-limiting toxicities observed at subtherapeutic levels[1][3][4][5].
