Overview

APS03118 is a next-generation, highly selective small molecule inhibitor of the RET (rearranged during transfection) tyrosine kinase. Developed by Applied Pharmaceutical Science (APS), it is designed to target a broad spectrum of RET alterations—including fusions and activating mutations such as gatekeeper (V804M/L/E) and solvent front (G810R/S/C) mutations—that confer resistance to first-generation RET inhibitors. Preclinical studies have demonstrated potent anti-tumor activity at nanomolar concentrations in both *in vitro* and *in vivo* models, including complete tumor regression in brain metastasis models with high survival rates. APS03118 has shown strong selectivity for RET over other kinases, suggesting a favorable safety profile. The drug has received IND approval and Fast Track Designation from the FDA; Phase 1 clinical trials are ongoing for patients with advanced solid tumors harboring RET alterations, including non-small cell lung cancer, thyroid cancer, pancreatic cancer, breast cancer, ovarian cancer, colorectal cancer, and other advanced malignancies[1][2][3][4][6][7][8].
