Overview
APTN-101 is a proprietary BRiTE (Brain Bispecific T cell Engager) therapeutic developed by Adaptin Bio for the treatment of glioblastoma (GBM), the most common and aggressive primary brain tumor. It is designed to target EGFRvIII, a mutant form of the epidermal growth factor receptor commonly found in malignant gliomas. The drug functions as a bispecific antibody that engages both EGFRvIII on tumor cells and CD3 on cytotoxic T lymphocytes, thereby stimulating immune-mediated cytotoxicity against cancer cells. Preclinical studies have demonstrated that APTN-101 achieves significantly enhanced distribution into the brain and shows potential for eradicating malignant glioma tumors. The FDA has cleared an Investigational New Drug application for APTN-101, enabling initiation of Phase 1 clinical trials in patients with WHO Grade IV Malignant Glioma (glioblastoma)[1][2][3][4][5][6][7].
