Overview

APVAC2 is a highly personalized peptide-based cancer vaccine designed for the treatment of glioblastoma. It consists of synthetic peptides that are unique to each patient's tumor, specifically targeting neoantigens arising from somatic, non-synonymous mutations found in the tumor. These mutated peptides are synthesized de novo for each individual and are not present in healthy tissue. The vaccine aims to stimulate the host immune system—primarily inducing CD4+ T helper cell type 1 (TH1) responses and some CD8+ cytotoxic T-cell responses—against tumor cells expressing these patient-specific mutated antigens, thereby reducing glioblastoma growth. APVAC2 is developed as part of a two-pillared strategy alongside APVAC1 (which targets shared non-mutated antigens), with both vaccines being tailored based on comprehensive analysis of the patient's tumor genome and immunopeptidome. The primary developers are BioNTech and Immatics N.V., with clinical evaluation conducted in newly diagnosed glioblastoma patients[1][3][4][5][7].
